Suspended within plasma, a single granulocyte is rendered in close-up with its surface membrane sharply defined against a defocused erythrocyte-rich background. The cytoplasm is packed with coarse, electron-dense specific granules, while a distinctly segmented nucleus occupies a central position, its lobes connected by thin chromatin bridges. Granules sit peripherally beneath the membrane and between nuclear lobes, consistent with an activated circulating leukocyte rather than a smear artifact. For teaching, this view is most often used to discriminate neutrophils from eosinophils and other leukocytes based on granule size, density, and nuclear morphology, a distinction that matters when you are correlating a differential count with clinical context. Neutrophils typically show finer, less intensely staining specific granules and a 2 to 5-lobed nucleus, while eosinophils trend toward bilobed nuclei with larger, more uniform granules; confusion here can blur discussions of acute bacterial infection and sepsis versus allergic disease, asthma, and helminth-associated eosinophilia.
