Description

Rendered as a single human mast cell with a spherical outline, the plasma membrane forms fine ruffles and short cytoplasmic projections that extend radially into the surrounding space. Numerous secretory granules cluster in the peripheral cytoplasm, with smaller granules scattered more centrally, while an oval to slightly indented nucleus sits eccentrically rather than in the geometric center. Granule density increases toward the cell cortex, a pattern consistent with regulated exocytosis at the membrane. Scale is implied at the cellular level rather than tissue architecture. Mast cells sit perivascularly in connective tissue and mucosa, and their granules contain histamine and heparin along with proteases such as tryptase, making them the effector cell for IgE mediated immediate hypersensitivity. Degranulation at the cell surface drives urticaria, bronchospasm, and anaphylaxis, and this kind of close peripheral packing helps learners connect receptor cross linking on the membrane to rapid mediator release into the interstitium. A useful teaching point is the distinction from circulating basophils (a granulocyte): basophils show a lobed nucleus and coarse basophilic granules on a blood smear, while mast cells are tissue resident and classically assessed with toluidine blue metachromasia or CD117 (KIT) immunostaining.
