Arising in a lateral view of the dorsal midbrain, the nucleus of the posterior commissure sits in the pretectal region near the superior colliculus, just rostral to the tectum and dorsal to the periaqueductal gray surrounding the cerebral aqueduct. As the sequence advances, the posterior commissure is traced coursing transversely across the roof of the aqueduct, with the nucleus positioned adjacent to its fibers at the dorsocaudal diencephalic junction. Neighboring landmarks such as the quadrigeminal plate, the pineal region, and the dorsal aspect of the midbrain tegmentum provide orientation as depth cues and subtle rotations clarify anterior to posterior relationships.
