Description
Retinal tissue appears in microscopic section with its layers stacked from the vitreous (internal) surface toward the choroid (external). Closest to the vitreous, the internal limiting membrane borders the nerve fiber layer and ganglion cell layer, followed by the inner plexiform layer where bipolar, amacrine, and ganglion cell processes synapse. Deeper, the inner nuclear layer transitions through the outer plexiform layer to the outer nuclear layer of photoreceptor nuclei, then the external limiting membrane, the photoreceptor layer (rods and cones, including inner and outer segments), and finally the retinal pigment epithelium at the outermost boundary. Ten layers. The vertical alignment makes the neural retina and pigment epithelium easy to separate conceptually. Layer-by-layer organization is where retinal pathology becomes legible: rhegmatogenous retinal detachment develops at the interface between the neurosensory retina and retinal pigment epithelium, while diabetic retinopathy and retinal vein occlusion map clinically to edema and hemorrhage affecting the inner retinal layers supplied by the central retinal artery. The clear separation of nuclear and plexiform strata supports teaching synaptic relays from photoreceptors to bipolar cells to ganglion cells, and it also helps explain why outer retinal disorders such as retinitis pigmentosa primarily compromise the outer nuclear layer and photoreceptor segments before inner retinal remodeling appears. Fine textural detail helps distinguish cell-dense laminae from synapse-rich plexiform zones.
