Beneath a semi-transparent lateral facial skin layer, the deep masseter is visualized on the lateral surface of the mandibular ramus, lying deep to the superficial masseter fibers and immediately inferior to the zygomatic arch. Superiorly, its fibers arise from the medial aspect of the zygomatic arch and descend to insert along the ramus, approaching the angle of the mandible inferiorly. The mandibular body sits anterior to the parotid region, while the external acoustic meatus and auricle provide a posterior landmark for the masticatory compartment. Bone guides the eye. Understanding the deep masseter matters when you need to localize pain and swelling at the angle of the jaw without confusing it with parotid pathology or temporomandibular joint disease. Bruxism and other parafunctional habits can hypertrophy the masseter, and in botulinum toxin injection planning the deep layer changes the apparent bulk and alters where injections risk diffusion toward the parotid duct and facial nerve branches. This perspective also supports teaching why the zygomatic arch functions as a hard superior boundary and why the mandibular ramus is the operative target during approaches to the masticator space.
